What are the commonly used Oligo purification methods and which applications are they suitable for?

Oligos can be purified using various methods, each with its own advantages and suitability for different applications:

1)  Desalting: Desalting is a simple purification method that removes small molecular weight impurities such as salts and unincorporated nucleotides. It is suitable for applications where high purity is not critical, such as some molecular biology applications like PCR amplification.

2)  Solid Phase Extraction (SPE): SPE involves binding oligos to a solid support, followed by washing to remove impurities and elution of purified oligos. It offers higher purity compared to desalting and is suitable for applications requiring higher purity, such as sequencing, cloning, and PCR.

3)  High Performance Liquid Chromatography (HPLC): HPLC separates oligos based on differences in their chemical properties, such as size, charge, and hydrophobicity. It provides high purity and is suitable for demanding applications such as oligonucleotide therapeutics, diagnostic assays, and structural studies.

4)  Polyacrylamide Gel Electrophoresis (PAGE): PAGE separates oligos based on their size by electrophoresis through a polyacrylamide gel matrix. It is often used for preparative purification of longer oligos (> 30 nucleotides) and is suitable for applications like sequencing, hybridization, and site-directed mutagenesis.

5)  Reverse-phase Chromatography (RPC): RPC separates oligos based on differences in their hydrophobicity. It is particularly useful for purifying modified oligos and is suitable for applications requiring high purity, such as RNA interference studies and antisense oligonucleotide therapeutics.

6)  Capillary Electrophoresis (CE): CE separates oligos based on differences in their charge-to-size ratio by electrophoresis through a capillary tube. It offers high resolution and is suitable for both analytical and preparative purification of oligos for various applications, including genomics, diagnostics, and gene synthesis.

Each purification method has its own advantages and limitations, and the choice of method depends on factors such as the required purity, yield, length of oligos, and specific application requirements.
